Will going from a original eguipment load range C light truck tire to a E rated tire of the same make and line have a fuel economy effect.

A tire with a higher load range is designed to be run at a higher inflation pressure. So, it could be heavier and have a higher rolling resistance value because of the extra weight and is constructed just a little different. But, even if there is a difference, in my opinion, you would never be able to measure the differance even if you track fuel economy very closely.
